Structural basis of salicylic acid perception by Arabidopsis NPR proteins

salicylic acid (SA) is a plant hormone that is essential for resistance to pathogens1-3. NPR protein has previously been identified as SA receptors4-10, although how they perceive SA and coordinate the hormonal signal remains unknown. Here we report the core mapping SA-binding Arabidopsis thaliana NPR4 and crystal structure of the ligand-bound her.

SA-binding core domain NPR4 folded with SA adopts α-helical fold that is actually buried in the hydrophobic core SA. Lack of ligand-entry path indicates that the SA binding conformation involves major renovation of the core-binding NPR4 SA, which we validated using mass spectrometry analysis of hydrogen-deuterium exchange full-length protein and through SA-induced disruption of interaction between NPR1 and NPR4. We show that, even though the two proteins share a nearly identical hormone-binding residues, NPR1 featuring minimal SA-binding activity compared to NPR4.

We identified a further two core surface residue SA-binding, mutations that alter the ability of the SA-binding and interaction with NPR1 NPR4. We also show that express a variant NPR4 hypersensitive to SA could increase the basal SA-mediated immunity without compromising the effector-triggered immunity, because this variant’s ability to re-associate with the high-level SA NPR1 remain intact. By revealing the structural mechanisms of perception SA by NPR proteins, our work paves the way for future investigations the specific role of this protein in SA signaling and their potential for engineering plant immunity.


Systemic acquired resistance (SAR) is an inducible defense mechanism that systematically increase resistance to pathogens in the leaf tissue. SAR, moving salicylic acid (SA) signaling, molecular component stocks with autonomous pathway, which is involved in controlling flowering time in Arabidopsis thaliana. FLOWERING LOCUS D (FLD) is one such autonomous path components needed for the time and the accumulation of systemic SA during flowering SAR. Here, we show that CYP720A1, a cytochrome P450 monooxygenases suspected, FLD expression and control required for flowering time and the manifestation of the SAR.

We are studying the genotype-phenotype map 80 quantitative phenotype in the model plant Arabidopsis thaliana, by representing genotypes affect each genotype phenotype as a network. In such networks, each corner or node corresponds to genotype individuals in all genomic loci that affect particular phenotypes.

Two vertices are connected by an edge if associated genotypes differ in exactly one nucleotide. 80 genotypes we analyze the network is based on data from genome-wide association study of 199 accessions of A. thaliana. They form a graph that connects the topography differ substantially between phenotype. We focus our analysis on the happenings epistasis (non-additive interaction between mutation) because of the high incidence of epistasis can reduce the accessibility of an evolutionary path toward phenotypic values ​​are high or low.

We find epistatic interactions in 67 phenotypes, and in 51 mutant phenotypes each pair is epistatic interactions. In addition, we found differences in phenotype-specific mutations in the fraction of accessible paths phenotypic maximum value. However, although epistasis affect accessibility phenotypic maximum value, the relationship between genotype and phenotype changes of our phenotypes analyzed subtle enough that some evolutionary pathway remains accessible to most phenotypes, even where epistasis pervasive.
