Deregulated High Affinity Copper Transport Alters Iron Homeostasis in Arabidopsis
This work describes the effects on iron homeostasis when copper transport was deregulated in Arabidopsis thaliana by overexpressing the high affinity copper transporter COPT1 and COPT3 (COPTOE). A genome-wide analysis carried out on the plant COPT1OE, highlighting that iron homeostasis affected the expression of genes under both copper deficiency and excess. Among the genes that change is their coding machine iron absorption and their transcriptional regulators. Furthermore, COPTOE seeds contained less iron and more sensitive than the control of iron deficiency.
Deregulation of copper (I) uptake inhibiting the transcriptional activation of Ib subgroup of basic helix-loop-helix (bHLH-Ib) factor under copper deficiency. In fact, excess copper inhibits the expression of master regulators FIT but activated bHLH-lb expression in plants COPTOE, in either case leading to a lack of response to an adequate iron absorption. Increase in media such as copper, iron (III) has been accumulated in the roots, and the ratio of iron (III) / iron (II) increases in crop COPTOE.
Thus, iron (III) COPTOE root overloading in iron deficiency inhibit the response of local, aims to metal uptake from the soil, leading to a general low iron content in seeds COPTOE. These results emphasize the importance of proper spatiotemporal copper absorption of iron homeostasis under non-optimal supply of copper. An understanding of the role of copper absorption in iron metabolism can be applied to improve plant resistance to iron deficiency.

We have previously reported that the expression level of polymorphism (ELP) from AtALMT1 strongly associated with variations in Al tolerance between natural Arabidopsis accessions.
However, although AtMATE also states follow Al exposure and contributed to Al tolerance, whether AtMATE contributes to the variation of Al tolerance and molecular mechanisms of the ELP is still unclear. Here, we dissect the natural variations in expression levels AtMATE in response to Al at the root using diverse natural accessions of Arabidopsis. Phylogenetic analysis showed that more than half of the population show accessions belonging to Central Asia (CA) real AtMATE low expression levels, while the majority of European populations show a high level of expression. Accession of the population AtMATE CA with low expression also showed significantly weakened Al tolerance.
A genome-wide association study of single-resident (GWAS) of AtMATE expression in CA identifying populations at AtMATE retrotransposon insertion of the promoter region of genes associated with a low level of expression. This can affect the transcriptional regulation of AtMATE by interfering with the effects of cis-regulatory elements located upstream of the insertion site, which include AtSTOP1 (sensitive to proton rhizotoxicity 1) transcription factor-binding sites revealed by the analysis of chromatin immunoprecipitation-qPCR.
